2.1 App Completeness

Purport to represent whole cities, regions, countries or official policy makers without the approval of DiEM25’s CC. For this purpose, they are assigned names such as DSC London 1, DSC Barcelona 1, etc. so that when other DSCs appear in the same vicinity they can adopt the names DSC London 2, DSC Barcelona 2 etc.

To make clear that opinions expressed are those of the particular DSC, and not of DiEM25 for the whole region/country in which the DSC is located, DSCs will use the white background logo for public online posts, while the broader organisation will use the orange/red logo. DSCs will also sign all press releases and other materials with their DSC name or with “DiEM25 Volunteers in …”, so as to distinguish them from messages by elected representatives of DiEM25. How to create a DSC

The up-to-date list of DSCs can always be found at www.diem25.org/local-groups-dscs/ . If there is already a DSC in your city, you are still encouraged to create another one, as each DSC is supposed to have no more than 15 active members. Of course you are encouraged to coordinate informally with the other DSCs in your area or group of DiEM25 members interested in the same theme/policy area.

Principles of successful DSC meetings

Small group discussion is the key element in empowering members. People become more committed through the physical act of speaking. Conversely if people do not speak at all in a meeting or very little, then they become passive and feel disempowered. The primary aim of DSC meetings should be to motivate and empower its members through peer-group activities that build a collective identity and a common political outlook.

DSCs must strive to find answers to the question: What do DiEM25 values and principles meanin practice?How can meetings make participants leave the (physical or digital) room feeling enthused rather than alienated?

DSCs are encouraged to discuss within them, but also across different DSCs, the rules of inclusive engagement so members are aware of them from the beginning, and to avoid having to conjure them up after anti-social and coercive behaviour is observed.

Procedure to create a local DSC

Go to the “Meet Local Members” forum and navigate to the subforum for your country or region. Either create or answer a post about forming a DSC there. Then write an email to volunteer@ null diem25.org . If you have already had a physical meeting of at least five people, you will receive an official DSC name and listing in the directory, where others can find you. If you haven’t yet had such a meeting, we can help you send a message to all DiEMers in your city in order to jump-start the process.

Insulin has recently also been shown to suppress AP-1 ( Aldo Felania Sneaker zbs7FlHCR
), a transcription factor that regulates the expression of matrix metalloproteinases (MMPs). MMPs are capable of lysing collagen and other matrix proteins and thereby allow the spread of inflammatory cells ( 33 36 . They thus mediate the thinning and finally the rupture of the atherosclerotic plaque, which triggers thrombosis as described above. MMPs also mediate the initial intimal injury, which allows the monocytes to enter the subendothelial intima in between endothelial cells. The suppression of MMPs secondary to that of AP-1 would therefore be protective to the endothelium and to the fibrous cap of the plaque. This could be useful in the clinical setting of unstable angina, which is associated with the rupture of the atherosclerotic plaque. AP-1 and MMPs are also suppressed by glucocorticoids in vitro ( 37 , 38 ) and in vivo ( Keen Newport Hydro Fisherman Sandal Womens 7RqtZVwE
). Glucocorticoids also suppress Egr-1 and, more than likely therefore, TF and PAI-1 ( 39 ).

One mechanism underlying the anti-inflammatory effect of insulin may be through the release of NO from the endothelium; NO is known to exert an anti-inflammatory effect ( 40 ). Insulin has also been shown to induce an increase in the expression of NO synthase (NOS), the enzyme that generates NO ( 41 ). The induction of NOS and the release of NO acutely impart to insulin a vasodilatory property through the action of NO on the vascular smooth muscle. A similar effect of insulin on platelets allows it to exert an anti-aggregatory effect. The platelet has NOS that is activated by insulin to induce the generation of NO. NO exerts its effects on vascular smooth muscle and platelets through the activation of guanylate cyclase, which generates cGMP from GTP ( Calvin Klein Mimette WoNAJHh
44 ). The vasodilatory and antiplatelet effects are clearly of importance in the treatment of acute myocardial infarction. It is also of interest that NO may mediate at least a part of the anti-inflammatory effect attributable to insulin since the coincubation of an NOS inhibitor, nitro-arginine, with insulin and human aortic endothelial cells neutralizes the suppressive effect of insulin on the expression of ICAM-1 by these cells ( Marc Jacobs Sheena Strap Sandal jL6vg

It should be mentioned that insulin sensitizers of the thiazolidinedione (TZD) class have been shown to exert anti-inflammatory effects including the suppression of NF-κB, ROS generation, and p47 phox subunit in mononuclear cell (MNC) and plasma concentrations of ICAM-1, MCP-1, TNF-α, and CRP. More recently, rosiglitazone has been shown to suppress plasma MMP-9 concentrations in diabetes ( 45 ). While one expects their effects to be useful in chronic situations, it is relevant that their anti-inflammatory effects are observed in 3–7 days in humans in vivo. In an experimental model of acute myocardial infarction, even a single dose of rosiglitazone has been shown to reduce myocardial damage by 50% ( 46 , 47 ). Therefore, the potential anti-inflammatory effect of TZDs in acute clinical situations requires investigation.
